Profiling reactive astrocyte subtypes at the molecular level


Characterizing the protein profiles of reactive astrocyte subtypes is crucial, given that proteins govern nearly all aspects of cell functions, including immunomodulation and metabolic signaling. In this context, I conducted proteomic profiling of a recently identified reactive astrocyte subtype termed "interferon-responsive reactive astrocytes" (IRRAs). My work identified over 2000 unique proteins within the cellular proteome, including several interferon-responsive proteins such as IFIT3, IFITM3, MX1, GBP2, and others. Additionally, I demonstrated the enrichment of IRRA markers in human AD postmortem brain tissue. 

In another collaborative effort, I also showed that a subset of astrocytes in human AD brains exhibit enrichment for neurotoxic reactive astrocyte-specific proteins. Importantly, these markers were identified using induced pluripotent stem cell-derived astrocytes in this study.
